While tadalafil (Cialis®) is widely used in clinical trials, the role of tadalafil in HC treatment is less well known and the role of sirolimus (Lymecycline®) in HC treatment is less well understood. In this study, we sought to evaluate the efficacy of tadalafil (Cialis®) and sirolimus (Lymecycline®) in patients with HC caused by a range of viruses, including HC caused by the same virus, and to evaluate the potential side effects of these drugs when used as monotherapy. We also sought to explore the use of tadalafil (Cialis®) and sirolimus (Lymecycline®) in combination in patients with HC who are not respond to the traditional combination therapy.
This is a retrospective study of patients who had a diagnosis of HC (including HC caused by the same virus, and the use of combination therapy) in a hospital in the UK between January 2020 and December 2021. The study protocol was reviewed and approved by the Ethics Committee of The Royal Free Hospital (Approval number: EU/11/037/001 and EU/11/037/002) in UK and the University of Birmingham (Approval number: MH/U11/0314). All patients signed a written consent form and met the study inclusion criteria.
Patients with HC caused by the same virus, HC caused by a different virus, HC caused by a combination therapy, or HC caused by a combination therapy were defined as patients who met the following inclusion criteria:
Patients with HC caused by HC viruses, HC caused by HC viruses with a similar virus and the use of combination therapy, and HC caused by HC viruses were included in the study.
In order to minimise the possibility of missing or unclear information on the timing of HC treatment, we used an open-label trial designed to evaluate the effectiveness of tadalafil (Cialis®) and sirolimus (Lymecycline®) in patients with HC caused by HC viruses.
Patients were treated with tadalafil (Cialis®) and sirolimus (Lymecycline®) as monotherapy and were then switched to either tadalafil (Cialis®) or sirolimus (Lymecycline®) in combination with a combination of tadalafil and sirolimus (Sirolimus®). Patients were given a combination of tadalafil and sirolimus in combination with a dose of 200 mg of tadalafil or a dose of 20 mg sirolimus, to ensure that both drugs had the same therapeutic effect. Blood samples for determination of HC virus load in the peripheral blood were collected at the time of treatment initiation for further analysis.
Patients who had an HC viral load of more than 3.0 × 106/mL (including HC caused by HC viruses, HC caused by HC viruses with a similar virus and the use of combination therapy) were included in the analysis.
